| GABAT

Gamma-amino-N-butyrate transaminase (GABAT) is a protein that in humans is encoded by the ABAT gene. This gene is located in chromosome 16 at position of 13.2. This gene goes by a number of names, including, GABA transaminase, GABAT, 4-aminobutyrate transaminase, NPD009 etc.It is mainly and abundant located in neuronal tissues.4-Aminobutyrate aminotransferase belongs to group of pyridoxal 5-phosphate-dependent enzyme which activates a large portion giving reaction to amino acids.ABAT is made up of two monomers of enzymes where each subunit has a molecular weight of 50kDa.It is identified that almost tierce of human synapses have GABA.GABA is a neurotransmitter that has different roles in different regions of the central and peripheral nervous systems. It can be found also in some tissues that do not have neurons.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 10 5 2.5 1.25 0.625 0.312 0.156 0
OD 2.26 1.45 0.85 0.55 0.36 0.27 0.19 0.07
Corrected OD 2.19 1.38 0.78 0.48 0.29 0.2 0.12 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Rat GABAT spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-101 96
Plasma(n=8) 92-105 103
Cell Lysates(n=8) 96-108 105

| Linearity

Samples were spiked with high concentrations of Rat GABAT and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Lysates(n=4)
1 : 2 Range (%) 84-95 88-96 90-110
Average (%) 90 93 96
1 : 4 Range (%) 89-103 87-108 105-113
Average (%) 94 98 107
