| DAF;CD55

Decay Accelerating Factor, also known as CD55 or DAF, is a protein that regulates the complement system on the cell surface. It recognizes C4b and C3b fragments, which are produced during the activation of C4 (classical or lectin pathway) or C3 (alternative pathway). It interacts with cell-associated C4b from both the classical and lectin pathways, interfering with the conversion of C2 to C2b and thereby preventing the formation of the C4b2a C3 convertase. It also interacts with C3b from the alternative pathway, interfering with the conversion of factor D to factor Bb by factor B, thus preventing the formation of the alternative pathway's C3bBb C3 convertase. Thus, by limiting the amplification of convertases in the complement cascade, DAF indirectly prevents the formation of membrane attack complexes. This glycoprotein is widely distributed in both hematopoietic and non-hematopoietic cells. It is a determinant of the Kromer blood group system.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (pg/mL) 350 175 87.5 43.75 21.87 10.93 5.46 0
OD 2.56 1.84 1.22 0.81 0.47 0.26 0.15 0.07
Corrected OD 2.49 1.77 1.15 0.74 0.4 0.19 0.08 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Mouse DAF;CD55 spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-102 96
Plasma(n=8) 93-105 101
Cell Culture Supernates(n=8) 96-107 102

| Linearity

Samples were spiked with high concentrations of Mouse DAF;CD55 and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Culture Supernates(n=4)
