| VCL

Vinculin is a 117-kDa cytoskeletal protein (1,066 amino acids) with an acidic N-terminal head and basic C-terminal tail connected by a proline-rich linker. The globular head domain binds talin and α-actinin and contains tyrosine phosphorylation sites (e.g., Y100, Y1065), while the tail interacts with F-actin, paxillin, and membrane lipids. This multidomain structure enables its role as a mechanical linker at focal adhesions and adherens junctions. Vinculin transitions between closed (auto-inhibited) and open (active) conformations to regulate cell adhesion, migration, and force transduction. Knockout models show embryonic lethality with cardiac and neural defects, underscoring its essential functions in development and tissue integrity.

| Recovery

The recovery of Mouse VCL spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

| Linearity
