Coagulation factor XII, also known as Hageman factor, is a plasma protein involved in coagulation. It is the zymogen form of factor XIIa , an enzyme of the serine protease (or serine endopeptidase) class. FXII is a plasma glycoprotein of approximately 9 kDa and is part of the coagulation cascade. In vivo, FXII is activated by binding to polyanions in a process termed contact activation. Activated platelets release inorganic polyphosphates, which activate FXII, initiating the intrinsic pathway and contributing to fibrin formation and thrombus development.| Typical data
As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.Concentration (ng/mL) | 100 | 50 | 25 | 12.5 | 6.25 | 3.12 | 1.56 | 0 |
OD | 2.6 | 1.9 | 1.24 | 0.75 | 0.46 | 0.31 | 0.22 | 0.07 |
Corrected OD | 2.53 | 1.83 | 1.17 | 0.68 | 0.39 | 0.24 | 0.15 | - |
| Repeatability
Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%| Recovery
The recovery of Human FⅫ spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.Sample Type | Range(%) | Average Recovery(%) |
Plasma(n=8) | 92-105 | 102 |
| Linearity
Samples were spiked with high concentrations of Human FⅫ and diluted with Universal Diluent to produce samples with values within the range of the assay.Plasma(n=4) | ||
