Aggrecan (ACAN), also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is a protein that is encoded by the ACAN gene. This gene is a member of the lectican (chondroitin sulfate proteoglycan) family. The encoded protein is an integral part of the extracellular matrix in cartilagenous tissue and it withstands compression in cartilage.Aggrecan is a proteoglycan, or a protein modified with large carbohydrates; the human form of the protein is 2316 amino acids long and can be expressed in multiple isoforms due to alternative splicing.Aggrecan was named for its ability to form large aggregates in the cartilage tissue (a large aggregating proteoglycan).| Typical data
As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.Concentration (ng/mL) | 30 | 15 | 7.5 | 3.75 | 1.87 | 0.93 | 0.46 | 0 |
OD | 2.48 | 1.31 | 0.93 | 0.52 | 0.33 | 0.22 | 0.16 | 0.07 |
Corrected OD | 2.41 | 1.24 | 0.86 | 0.45 | 0.26 | 0.15 | 0.09 | - |
| Repeatability
Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%| Recovery
The recovery of Human AGC spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.Sample Type | Range(%) | Average Recovery(%) |
Serum(n=8) | 84-101 | 97 |
Plasma(n=8) | 92-105 | 102 |
Cell Culture Supernates(n=8) | 98-108 | 105 |
| Linearity
Samples were spiked with high concentrations of Human AGC and diluted with Universal Diluent to produce samples with values within the range of the assay.Serum(n=4) | Plasma(n=4) | Cell Culture Supernates(n=4) | ||
1 : 2 | Range (%) | 84-95 | 88-96 | 90-110 |
