| sOX40L

OX40L is the ligand for OX40 and is stably expressed on many antigen-presenting cells such as DC2s (a subtype of dendritic cells),macrophages, and activated B lymphocytes. The ligation of OX40-OX40L is a source of survival signal for T cells[6] and enables the development of memory T cells.Signaling through these two molecules also leads to polarization towards Th2 immune response even in an environment with low levels of IL-4 cytokine.OX40L is also present on the surface of many non-immune cells, for example, endothelial cells and smooth muscle cells. The surface expression of OX40L is induced by many pro-inflammatory mediators, such as TNF-α, e.g. produced by mast cells, IFN-γ and PGE2 (prostaglandin E2).OX40L has also been designated CD252 (cluster of differentiation 252).

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 10 5 2.5 1.25 0.625 0.312 0.156 0
OD 2.52 1.96 1.38 0.97 0.55 0.31 0.19 0.08
Corrected OD 2.44 1.88 1.3 0.88 0.47 0.23 0.11 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human sOX40L spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Tissue Homogenates(n=8) 92-105 102
Cell Lysates(n=8) 96-108 105

| Linearity

Samples were spiked with high concentrations of Human sOX40L and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Tissue Homogenates(n=4) Cell Lysates(n=4)
1 : 2 Range (%) 88-100 90-110
Average (%) 93 96
1 : 4 Range (%) 87-107 105-115
Average (%) 98 109
