| PCSK5

Proprotein convertase subtilisin/kexin type 5 is an enzyme that is encoded by the PCSK5 gene, found in chromosome 9q21.3 .Two alternatively spliced transcripts are described for this gene but only one has its full length nature known. The protein encoded by this gene belongs to the subtilisin-like proprotein convertase family. The members of this family are proprotein convertases that process latent precursor proteins into their biologically active products. This encoded protein mediates posttranslational endoproteolytic processing for several integrin alpha subunits. It is thought to process prorenin, pro-membrane type-1 matrix metalloproteinase and HIV-1 glycoprotein gp160.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 10 5 2.5 1.25 0.625 0.312 0.156 0
OD 2.27 1.62 1.07 0.65 0.44 0.33 0.24 0.08
Corrected OD 2.19 1.54 0.99 0.57 0.36 0.25 0.16 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human PCSK5 spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Tissue Homogenates(n=8) 86-101 96
Cell Culture Supernates(n=8) 92-105 103

| Linearity

Samples were spiked with high concentrations of Human PCSK5 and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Tissue Homogenates(n=4) Cell Culture Supernates(n=4)
1 : 2 Range (%) 84-95 88-98
Average (%) 91 93
1 : 4 Range (%) 89-103 87-108
Average (%) 95 98
