| NCCT

The sodium-chloride symporter (also known as Na+-Cl− cotransporter, NCC or NCCT, or as the thiazide-sensitive Na+-Cl− cotransporter or TSC) is a cotransporter in the kidney which has the function of reabsorbing sodium and chloride ions from the tubular fluid into the cells of the distal convoluted tubule of the nephron. It is a member of the SLC12 cotransporter family of electroneutral cation-coupled chloride cotransporters. A loss of NCC function causes Gitelman syndrome, an autosomic recessive disease characterized by salt wasting and low blood pressure, hypokalemic metabolic alkalosis, hypomagnesemia and hypocalciuria.Over a hundred different mutations in the NCC gene have been identified.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 20 10 5 2.5 1.25 0.625 0.312 0
OD 2.44 1.91 1.37 0.84 0.51 0.34 0.26 0.07
Corrected OD 2.37 1.84 1.3 0.77 0.44 0.27 0.19 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human NCCT spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Tissue Homogenates(n=8) 84-102 96
Cell Lysates(n=8) 92-105 100

| Linearity

Samples were spiked with high concentrations of Human NCCT and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Tissue Homogenates(n=4) Cell Lysates(n=4)
1 : 2 Range (%) 82-95 88-96
Average (%) 91 93
1 : 4 Range (%) 89-103 87-108
Average (%) 94 100
