| alpha-Synuclein

Alpha-synuclein (aSyn) is a protein that is encoded by the SNCA gene.Alpha-synuclein is a neuronal protein that regulates synaptic vesicle trafficking and subsequent neurotransmitter release. It is abundant in the brain, while smaller amounts are found in the heart, muscle and other tissues. In the brain, alpha-synuclein is found mainly in the axon terminals of presynaptic neurons.Within these terminals, alpha-synuclein interacts with phospholipids and proteins.Presynaptic terminals release chemical messengers, called neurotransmitters, from compartments known as synaptic vesicles. The release of neurotransmitters relays signals between neurons and is critical for normal brain function.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 20 10 5 2.5 1.25 0.625 0.312 0
OD 2.107 1.46 0.984 0.685 0.44 0.319 0.236 0.135
Corrected OD 1.972 1.325 0.849 0.55 0.305 0.184 0.101 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human alpha-Synuclein spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 82-102 97
Plasma(n=8) 91-104 103
Cell Culture Supernates(n=8) 95-108 102

| Linearity

Samples were spiked with high concentrations of Human alpha-Synuclein and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Culture Supernates(n=4)
1 : 2 Range (%) 84-94 84-96 91-110
Average (%) 92 91 95
1 : 4 Range (%) 89-102 97-108 105-114
Average (%) 93 97 105
