Alkaline phosphatase, placental type also known as placental alkaline phosphatase (PLAP) is an allosteric enzyme encoded by the ALPP gene. This gene is polymorphic and three common alleles (type 1, type 2, and type 3) for this form of alkaline phosphatase have been well-characterized. Alkaline phosphatase, placental type is a membrane-bound glycosylated dimeric enzyme, also referred to as the heat-stable form, that is expressed primarily in the placenta, although it is closely related to the intestinal form of the enzyme as well as to the placental-like form.| Typical data
As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.Concentration (pg/mL) | 800 | 400 | 200 | 100 | 50 | 25 | 12.5 | 0 |
OD | 2.41 | 1.84 | 1.21 | 0.85 | 0.52 | 0.33 | 0.21 | 0.08 |
Corrected OD | 2.33 | 1.76 | 1.13 | 0.77 | 0.44 | 0.25 | 0.13 | - |
| Repeatability
Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%| Recovery
The recovery of Human PLAP spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.Sample Type | Range(%) | Average Recovery(%) |
Serum(n=8) | 84-101 | 96 |
Plasma(n=8) | 92-105 | 102 |
Tissue Homogenates(n=8) | 96-108 | 105 |
| Linearity
Samples were spiked with high concentrations of Human PLAP and diluted with Universal Diluent to produce samples with values within the range of the assay.Serum(n=4) | Plasma(n=4) | Tissue Homogenates(n=4) | ||
